Целью работы явилась разработка эмульсионной трансдермальной терапевтической системы (ТТС) циклоспорина и исследование диффузии лекарственного вещества из ТТС.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Количественное определение циклоспорина в модельных средах проводили на жидкостном хроматографе Agilent 1100 G1311 со спектрофотометрическим детектором G1315B (Германия). Для изготовления эмульсионной матрицы использовали диспергатор Heidolph DIAX900 (Германия) и ультразвуковой гомогенизатор Heilscher UIS250V (Германия). Исследования диффузии циклоспорина из ТТС через мембрану Strat-M (25 мм в диаметре, Merck Millipore) проводили на анализаторе диффузии Copley (Великобритания).</p></sec><sec><title>Результаты</title><p>Результаты. С помощью разработанного метода высокоэффективной жидкостной хроматографии установлено, что количество циклоспорина, прошедшего за 24 часа через мембрану Strat-M из ТТС размером 10 см2, содержащей 173 мг субстанции, составило 5,9 мг. При этом расчетная концентрация в крови составляет ~ 50 нг/мл, что соответствует терапевтической поддерживающей концентрации в крови пациентов (50–75 нг/мл) после трансплантации органов.</p></sec><sec><title>Заключение</title><p>Заключение. В модельных экспериментах показана принципиальная возможность трансдермального переноса циклоспорина.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Aim</title><p>Aim: to develop the emulsion transdermal therapeutic system (TTS) of cyclosporine and the research of its diffusion from TTS.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The quantitative evaluation of cyclosporine in model media was performed on liquid chromatograph Agilent 1100 G1311 with spectrophotometric detector G1315B (Germany). For the production of the emulsion matrix, the Heidolph DIAX900 disperser (Germany) and the ultrasonic homogenizer Hielscher UIS250V (Germany) were used. The cyclosporine diffusion from TTS through Strat-M membrane (25 mm in diameter, Merck Millipore) was studied using Copley diffusion analyser (Great Britain).</p></sec><sec><title>Results</title><p>Results. Using the developed method of micro-HPLC, it was established that the amount of cyclosporine passed through the Strat-M membrane over 24 hours from 10 sm2 TTS, containing 173 mg of the substance, was 5.9 mg. The estimated blood concentration is ~ 50 ng/ml which corresponds to the therapeutic maintenance blood concentration during organ transplantation (50–75 ng/ml).</p></sec><sec><title>Conclusion</title><p>Conclusion.
